Core Tip Analyzing the very low abundance of proteins contained in small extracellular vesicles is technically challenging, particularly when proteins are further separated into membrane and lumenal fractions. This review describes a practical method for fractionating small extracellular vesicles into membrane and lumenal components and introduces an ultrasensitive assay capable of quantifying trace amounts of proteins in each fraction through the combined use of commercially available kits.
