Core Tip This study examines the metabolic significance of elevated gonadotropin-releasing hormone (GnRH) in the postmenopausal state. We found that GnRH receptor is preferentially expressed in pancreatic β-cells and that GnRH enhances insulin secretion through the cAMP-PKA-PDX1 signaling pathway. These findings suggest that postmenopausal hypothalamic-pituitary-gonadal axis remodeling may contribute to β-cell compensation and provide a mechanistic link between reproductive endocrine aging and glucose homeostasis.
