| Core Tip |
Colorectal cancer (CRC) is a leading cause of cancer-related mortality. Glutamine (Gln) metabolism and mechanistic target of rapamycin complex 1 (mTORC1) signaling are crucial for tumor growth. This study reveals that activating transcription factor 4 (ATF4) directly upregulates the Gln transporter alanine-serine-cysteine transporter 2, which mediates Gln uptake, activates mTORC1 signaling, and promotes CRC proliferation. Our findings are consistent with a proposed ATF4-alanine-serine-cysteine transporter 2-mTORC1 positive feedback loop in CRC, which requires direct functional validation of the reciprocal mTORC1/URB1→ATF4 arm. |
