Core Tip Gan-tang-yi decoction (GTYD) alleviates liver injury, fibrosis, and insulin resistance in cirrhotic rats by suppressing endoplasmic reticulum stress. GTYD inhibits the pro-apoptotic PERK/ATF4/CHOP signaling pathway both in vivo and in vitro. Activation of PERK by CCT020312 reverses the hepatoprotective effects of GTYD, confirming its pathway specificity. GTYD reduces carbon tetrachloride-induced hepatocyte apoptosis by attenuating endoplasmic reticulum stress, as validated by the effects of 4-phenyl butyrate and thapsigargin.
