Core Tip Abnormal migration of enteric neural crest cells is a major cause of intestinal developmental abnormalities. This study characterized PDLMNA, an endogenous peptide that binds to α-ketoglutarate-dependent demethylase 5 (ALKBH5) and disrupts the interaction between ALKBH5 and Lamin A/C. PDLMNA effectively restores impaired enteric neural crest cells migration by modulating the LMNA-ALKBH5 complex, revealing a novel peptide-mediated molecular mechanism involved in enteric nervous system development.
