| Core Tip |
This retrospective, single-center cohort study evaluated the prognostic relevance of molecular alterations detected by next-generation sequencing in 208 patients with metastatic colorectal carcinoma (CRC). BRAF mutations were more frequent in right-sided tumors and were associated with shorter overall survival (OS) in the Kaplan-Meier analysis but were not associated with OS after multivariable adjustment. The FBXW7 mutation, although infrequent, was associated with shorter OS after adjustment for the available covariates. These findings provide hypothesis-generating support for the prognostic relevance of FBXW7 mutation in metastatic CRC and extend previous mutation-based observations, while underscoring the need for validation in larger, clinically well-characterized cohorts. |
