| Core Tip |
Hepatitis B surface antigen persistence is the primary driver of immune tolerance in chronic hepatitis B. Strategies that reduce or eliminate hepatitis B surface antigen to break host immune tolerance and reinvigorate an immune response are emerging as promising therapeutic avenues for clinical cure. These therapeutic modalities include monoclonal antibodies, small interfering RNAs, antisense oligonucleotides, gene-editing technologies, capsid assembly modulators, agents targeting secretory pathways, and immune-based clearance strategies. Combination strategies have shown synergistic effects. Future clinical trial designs must prioritize identifying optimal combinations that maximize synergy while minimizing toxicity. |
