| Core Tip |
This study presents a comprehensive transcriptomic framework to evaluate steatosis-targeting therapies by analyzing steatosis-related gene modulation, hepatocyte identity, cellular stress, and donor consistency, going beyond lipid reduction alone. Among the five therapies studied, diacylglycerol acyltransferase 2 inhibition demonstrated the most favorable relative exploratory profile. We further explored the association of the diacylglycerol acyltransferase 2 inhibition-derived signature with the transcriptional state of hepatocytes by exploratory single-cell analysis. However, these results remain hypothesis-generating due to the limited number of donors, the post hoc selection of signatures, and the pseudoreplication of single cells. |
