Core Tip This review reframes glyceraldehyde-3-phosphate dehydrogenase (GAPDH) as a multifunctional oncogenic hub, not just a glycolytic enzyme. Beyond metabolism, GAPDH stabilizes oncogene mRNAs (cyclin D1/c-Myc), activates hypoxia-inducible factor-1alpha, and drives metastasis, chemoresistance, and stemness. Targeting both its enzymatic and moonlighting roles with multi-action inhibitors (e.g., koningic acid, alternol) offers a strategic approach to exploit cancer-specific dependencies, simultaneously impairing energy production, survival, and treatment resistance despite GAPDH’s ubiquitous expression.
