This study integrates two-sample Mendelian randomization with tissue- and cell-type-specific expression and methylation quantitative trait loci to investigate shared genetic features between insomnia and ten psychiatric disorders. Genetically predicted insomnia was associated with bipolar disorder, major depressive disorder, and neuroticism after multiple-testing correction. Multi-omics analyses highlighted excitatory neurons and identified overlapping methylation–expression signals, including those involving MAP2K5 and CLP1. Integration with drug-target data further identified 14 candidate targets. These findings provide testable molecular hypotheses but require functional and clinical validation. |
