| Core Tip |
Ferroptosis is a form of cell death driven by iron-dependent lipid peroxidation, and its regulation involves iron metabolism, lipid remodeling, and glutathione peroxidase 4-centered antioxidant systems. In colorectal cancer cells, this process is further modulated by the signaling pathways of acyl-CoA synthetase long-chain family member 4, AMP-activated protein kinase, and nuclear factor erythroid 2-related factor 2. Ferroptosis reshapes the immunosuppressive tumor microenvironment through damage-associated molecular patterns-mediated immunogenic cell death, enhancing CD8+ T cell-mediated anti-tumor immunity; however, excessive lipid peroxides can compromise effector T cell viability. Ferroptosis-inducing combination therapy potently overcomes resistance to chemotherapies, targeted agents, and immune checkpoint inhibitors. Key translational challenges include tumor heterogeneity, the lack of ferroptosis-specific biomarkers, and off-target intestinal toxicity, which may be partially addressed through tumor-targeted nanomedicine and multi-omics patient stratification. |
