Core Tip This study evaluated the protective effects and mechanism of Weiduqing formula in gastric precancerous lesions. In rat and gastric epithelial cell models induced by N-methyl-N’-nitro-N-nitrosoguanidine, treatment reduced mucosal atrophy, intestinal metaplasia, inflammation, and malignant phenotypic changes. Integrated serum pharmacochemistry, metabolomics, proteomics, and network pharmacology prioritized stearoyl-CoA desaturase 1 (SCD1) as a treatment-responsive mediator. Functional experiments showed restored autophagic flux and an autophagy-related protein 5-dependent increase in SCD1-p62 co-localization. Weiduqing formula also partially remodeled gastric microbial composition. These findings identify a candidate mechanism and support further translational study.
