Core Tip Emerging histone post-translational modifications (HPTMs) connect metabolic reprogramming with epigenetic regulation in colorectal cancer (CRC). This review critically examines six emerging HPTMs – lactylation, crotonylation, citrullination, succinylation, propionylation, and monoaminylation – with emphasis on their roles in tumor progression, immune regulation, metastasis, and treatment resistance. Importantly, we distinguish direct human CRC evidence from CRC experimental, other-cancer, and noncancer evidence. We further discuss the emerging crosstalk linking HPTMs with epigenetic and metabolic regulation, together with advances in HPTM detection and potential therapeutic opportunities. Current evidence remains heterogeneous and predominantly preclinical, underscoring the need for further mechanistic and clinical validation.
