Core Tip Pancreatic ductal adenocarcinoma is associated with distinct biliary microbial and metabolic signatures that enable discriminative diagnosis. Depletion of microbiota-associated α-tocopherol (α-Toc) is linked to tumorigenesis. Mechanistically, α-Toc activates G protein subunit γ10, dephosphorylates phosphoinositide 3-kinase and AKT, induces p21, and triggers G0/G1 cell cycle arrest, thereby suppressing pancreatic ductal adenocarcinoma oncogenesis. All these effects are fully abolished upon G protein subunit γ10 silencing. Restoration of α-Toc reactivates this tumor-suppressive pathway, supporting its role as a candidate dual diagnostic-therapeutic axis.
