This study delves into the role of chaperonin-containing tailless complex polypeptide 1 subunit 6A (CCT6A), a protein folding chaperone, in CRC therapy. Our findings reveal that CCT6A expression is downregulated by cisplatin (DDP) in CRC cells, enhancing DDP's cytotoxicity. Depletion of CCT6A further potentiates DDP's effects by promoting autophagy, apoptosis, and necroptosis, while its overexpression provides resistance. Using immunoblotting and autophagy assays, we confirmed CCT6A's negative regulatory role in autophagy. Markers of apoptosis and necroptosis were upregulated in CCT6A-depleted cells, indicating increased cell death. Autophagy inhibition exacerbated cell death, compromising viability and proliferation. Thus, CCT6A emerges as a potential therapeutic target in CRC, modulating key cellular processes and enhancing DDP efficacy. |
