Cholangiocarcinoma (CCA) is a highly lethal malignancy of the biliary tract with very few treatment options. Novel biomarkers are urgently needed for CCA management and treatment. The novelties of this study are as follows: (1) our study, for the first time, showed that high-mobility group box 1 (HMGB1) is an independent prognostic factor in IHCC and positively correlate with lymphatic microvessel density and vascular endothelial growth factor C (VEGF-C) expressions; and (2) in vitro analysis suggested that HMGB1 enhances the secretion of VEGF-C and promotes Epithelial-mesenchymal transition of RBE cells. In all, our results define an important role of HMGB1 in the progression of cholangiocarcinoma, and HMGB1 may serve as a prognostic marker for IHCC patients.
