In this review, we summary the association between the PNPLA3 I148M variant and nonalcoholic fatty liver disease (NAFLD), and especially its role in nonalcoholic fatty liver fibrosis. The variant is associated with NAFLD, but is predominant in women, not in men. The association may vary among different ethnic populations, but is not affected by the presence of metabolic syndrome. Then, we speculate a pro-fibrogenic network that the PNPLA3 I148M variant may promote the development of fibrogenesis by activating the Hh signaling pathway, which, in turn, leads to the activation and proliferation of hepatic stellate cells, and excessive generation and deposition of extracellular matrix. |
