We studied whether in vivo and in vitro effects of granisetron might be influenced, at least in part, by the heme oxygenase/carbon monoxide (HO/CO) pathway. Our findings demonstrate for the first time that the HO/CO pathway takes part in the contractile colon activity in rats. Interestingly, the constipating effects of granisetron are positively correlated with levels of carbon monoxide, thus suggesting that treatments able to modulate carbon monoxide levels may potentially reduce the constipation mediated by granisetron. |
