In this study, we explored the role of the p38 MAPK pathway in the TNF-α-induced inflammation and apoptosis of pulmonary microvascular endothelial cell (PMVEC) in vitro and verified the effect of SB203580-mediated p38 inhibition on severe acute pancreatitis-related acute lung injury (SAP-ALI) in rats. We found that p38 MAPK overactivation promoted TNF-α-induced inflammatory cytokine expression and PMVECs apoptosis in vitro. Furthermore, SB203580 improved SAP-ALI by downregulating the levels of p-p38, NFκB, p-SATA3 and Myd88 in the lung. On the other side, it upregulated the IκB and HO-1 Levels in the lungs, which alleviated inflammation and apoptotic cell death in rats. |
