Core Tip This study demonstrated that the potential of hypoxic condition on human placenta-derived mesenchymal stem cells (hP-MSCs) to enhance the proliferation capacity. Through the use of RNA sequencing assays and molecular biology experiments, a novel gene CD99 was found to be involved in hypoxia-mediated promotion of hP-MSCs proliferation. Furthermore, it indicated that CD99 activates pathways that transport ERK1/2 to the nucleus, increasing the activity of cell cycle-associated proteins. Accordingly, hypoxia-inducible factor 1α-mediated CD99 plays a role in proliferation by regulating the MAPK/ERK signaling pathway.
