18β-glycyrrhetinic acid (18β-GRA) is an important bioactive component of glycyrrhiza liquorice. Our results showed that 18β-GRA up-regulates the expression of miR-328-3p in gastric cancer (GC) cell. Overexpression of miR-328-3p inhibits GC cell proliferation and colony formation, arrests the cell cycle, promotes apoptosis, inhibits subcutaneous tumor formation, and signal transducer and activator of transcription 3 (STAT3) expression. Dual-luciferase reporter assay showed that miR-328-3p could target the regulation of STAT3. The study of monomeric Red Fluorescent Protein-green fluorescent protein-light chain 3 double-labeled adenovirus and western blot indicated that 18β-GRA promotes autophagy flow by promoting autophagosome synthesis. These data suggest that 18β-GRA promotes cell autophagy and inhibits the proliferation of GC cell by regulating miR-328-3p/STAT3. |
